Bacteria are exquisite living machines that could one day deliver medicine to cells in the human body, if scientists overcome the numerous obstacles

Tiny robots that swim through our blood vessels attacking viruses and malignant cells have not quite crossed the line that separates science fiction from science?but there might be a way to jump-start their development.
Engineering nimble robots that are smaller than blood cells is extremely challenging. Rather than design them from scratch, some scientists have been experimenting with the idea of enlisting an army of sophisticated nanobots already at our disposal: the thousands of species of bacteria swarming inside our bodies right now. In recent years researchers have saddled microorganisms with useful nanoparticles and bits of DNA. Although the research is preliminary, some engineers and microbiologists see realizable potential. This week, at the American Chemical Society's annual meeting in San Diego, biomolecular engineer David Gracias of Johns Hopkins University discussed the progress he and his colleagues have made in gluing nanoparticles to bacteria.
So far, Gracias and his colleagues have managed to decorate Escherichia coli with incredibly tiny beads, rods and crescents made from nickel and tin coated in gold. Each nanoparticle is about 300 nanometers in diamter, or three times the size of a particle of wood smoke. The researchers stuck the nanoparticles to the bacteria with antibodies, which are small, Y-shaped proteins that the immune system uses to identify foreign cells, viruses and harmful bacteria. Each antibody fits like a puzzle piece into an array of different proteins on the surfaces of invading pathogens; antibodies can also lock onto other antibodies?usually from a different species. Gracias and his colleagues first coated the nanoparticles with antibodies from goats and rabbits, and subsequently coated the bacteria in complementary antibodies. When they soaked the bacteria in a solution of the nanoparticles and heated the mixture, the beads, rods and crescents stuck to the bacteria, like so many Lucky Charms marshmallows clinging to the underside of a wet spoon.
Gracias and his teammates also devised a way to load bacteria with nanoparticles at tiny docking stations and release them on demand. First they covered small squares of silicon and gold with a field of Y-shaped antibodies, to which they attached small beads dotted with complementary antibodies. Then they introduced bacteria sporting another set of complementary antibodies to the awaiting beads?as expected, they clasped like Velcro. A wave of dissolving chemicals detached the bacteria and their cargo beads from the docking station.
Many bacteria saddled with nanoparticles were still able to move around freely, albeit not as quickly as their unencumbered brethren. Sometimes, though, the backpacking bacteria just spun in circles, apparently unable to move forward. "This is definitely a work in progress," Gracias says. "Right now we are mostly focusing on varying the size and shape of the nanoparticles and making sure they stick."
Such nanoparticles can be heated from afar with infrared light, destroying diseased tissue. Ultimately, Gracias dreams of coaxing bacteria to ferry spongy nanoparticles soaked in drugs and outfitting bacteria with tiny sensors that measure local temperature and pH or tiny tools that perform surgery on a single cell. Similar research by other scientists confirms the potential of engineered bacteria to deliver medical packages directly into living cells.
In earlier work, Demir Akin of Stanford University and his colleagues used antibodies and nanoparticles to attach molecules of DNA to weakened Listeria monocytogenes, which is a bacterium responsible for many cases of food poisoning. L. monocytogenes is an intracellular bacterium, which means it has evolved ways to get inside animal cells. Akin attached a luciferase gene?which codes for the enzyme that makes fireflies glow?to L. monocytogenes and injected the germs into living mice. Three days later the mice glowed under a specialized camera, confirming that not only had the bacteria entered the mouse's cells, the cell nuclei had incorporated the bacteria's cargo and expressed the gene. Akin designed the living microbots to release their DNA packages inside mammalian cells, where the pH is low (acidic) enough to dissolve the chemical bonds gluing the luciferase gene to the bacteria.
The advantage of L. monocytogenes is that it knows how to get into cells, but it would be risky to use even a weakened version as a medical workhorse because it makes people sick. E. coli, in contrast, are much more harmless, but not all strains have specific adaptations for entering cells. The key, says Douglas Weibel of the University of Wisconsin?Madison, is working with a harmless microorganism that is a strong swimmer and has no problem butting its way into mammalian cells. In an experiment they conducted largely for fun, Weibel, George Whitesides of Harvard University and their colleagues yoked nanosize polystyrene beads to a single-cell green alga called Chlamydomonas reinhardtii. Weibel and Whitesides successfully steered their "microoxen" by shining light on one side of the algal cells (algae move toward light).
After that, Weibel did not pursue the challenge of engineering microorganisms to ferry nanoparticles, but he remains fascinated by the ongoing research. "Bacteria have already figured out how to move around in the body," he says. "They have evolved amazing motility. They can sense changes in their environment and adapt, not only on a short timescale, but they can adapt genetically, too. Even if we can't get them to deliver things in the human body, they could be useful for transporting nanoparticles in the lab. Who knows what advances we'll have 50 years from now."

Syria's army resumed heavy shelling of the rebel hubs of Homs and Hama on Saturday, monitors reported, as international envoy Kofi Annan arrived in Russia in a new push for peace.
Mortar rounds rained down on the flashpoint central city of Homs and nearby town of Qusayr from early morning, killing at least 10 people, said the Syrian Observatory for Human Rights.
At least 20 people were reported killed nationwide, among them four government soldiers.
The fiercest regime assault targeted Saraqeb in the northwestern province of Idlib which was stormed by troops backed by 26 tanks that "took up position to split the town in two," activist Nureddin al-Abdo told AFP.
Residents cowered indoors as explosions rattled Saraqeb and troops swooped to make arrests, he said from the town where there is a considerable rebel Free Syrian Army (FSA) presence.
One civilian and three soldiers died in Saraqeb, the Observatory said.
The army also used mortars to pound the town of Qalaat al-Madiq in the central province of Hama, which troops have been trying to seize for the past two weeks, added the Britain-based monitoring group.
Activists reported fierce overnight clashes between troops and mutineers in and around the capital, after a day of protests under the rallying cry "Damascus, here we come."

A top Kremlin-linked lawmaker said Assad should treat the UN statement as "an insistent recommendation" whose implementation would determine the future course of relations between the two countries.

Odd Future have some pretty famous fans. The band of misfits has won over some unlikely pop stars, including Willow Smith and Justin Bieber, as well as rap royalty like Kanye West and Lil Wayne.
In an October interview, Tyler, the Creator revealed to MTV News that he wanted to make a '90s-inspired R&B track with break beats and hard drums for Bieber, but he refused to give up any more info when he appeared on Wednesday's "RapFix Live."
"That's my f---ing dawg. Shout-out to Justin," Tyler said.
When asked about the dream collaboration, the rap star only offered up: "You'll see."
In the past year, Tyler has formed a bond with musical idol Pharrell Williams, Lil Wayne and Kanye West and even interviewed Nas for an XXL magazine cover story.
"It's cool. That sh-- is cool. Awkward, but it's tight. They actually f--- with us," Tyler said of all his new friends. "Lil Wayne is actually a fan, Kanye is actually like a fan. I actually talk to those dudes all the time."
Taco said he and Wayne usually talk about skating, and Earl began to recall a phone conversation between him and Weezy before he was made fun of by the rest of the Wolf Gang. "The moment is gone now, but I just kept saying 'no' and it was awkward," Earl squealed.
"We don't really talk about anything deep," Tyler said. "I don't know, I don't really sit there and say, 'Damn, what conversation are me and Wayne having right now?' "

Prostate Cancer Research Program?Idea Development Awards

SYNOPSIS: The overall goal of the FY12 PCRP is to find and fund research that will lead to the elimination of death and suffering from prostate cancer. Specifically, the PCRP seeks to promote highly innovative, groundbreaking research; high-impact research with near-term clinical relevance; multidisciplinary synergistic research; translational studies to support the fluid transfer of knowledge between bedside and bench; research on patient survivorship and quality of life; the next generation of prostate cancer investigators through mentored research; and research on disparities in the incidence and mortality of prostate cancer. The FY12 Idea Development Award mechanism encourages applications from investigators in the early stages of their careers. The New Investigator Option is designed to allow applicants early in their faculty appointments, or in the process of developing independent research careers, to compete for funding separately from established investigators. Applications from New Investigators and Established Investigators will be peer- and programmatically reviewed in separate groups. PIs using the New Investigator Option are strongly encouraged to strengthen their applications by collaborating with investigators experienced in prostate cancer research and/or possessing other relevant expertise. It is the responsibility of the applicant to describe how collaboration will augment the PIs expertise to best address the research question. All applicants for the New Investigator Option must meet specific eligibility criteria.

WASHINGTON (AP) ? Mitt Romney says he's "almost there" in his six-year pursuit of the Republican presidential nomination after defeating Rick Santorum in the Illinois GOP primary. There are signs that big GOP donors agree, as major contributors shift more dollars in his direction.
"Tonight's win means we are that much closer to securing the nomination, uniting our party, and taking on President Obama," the Republican front-runner wrote in a campaign email sent late Tuesday.
He urged the party to fall in line behind his bid, saying, "We are almost there."
The former Massachusetts governor and his allies spent hundreds of thousands of dollars more than Santorum and his backers in Illinois, and it showed in the results: Romney was beating Santorum by 47 percent to 35 percent.
Campaign finance reports released Tuesday showed that big donors to a GOP political organization founded by political strategist Karl Rove have boosted their financial support for Romney in recent weeks.
For all of that money, though, Romney's Illinois win was a victory without an electrified electorate: Turnout seemed likely to be among the lowest in decades: Officials in several election districts said turnout hovered around 20 percent.
"You could draw a bigger crowd at a Green Bay Packers rally in downtown Chicago than what Mr. Romney delivered to the polls," Democratic Sen. Dick Durbin of Illinois said on "CBS This Morning."
Romney was the clear favorite among Illinois Republicans who were most concerned about picking someone who is capable of taking on President Barack Obama in the fall. Romney's wife, Ann, suggested earlier this week that it was time for the party to coalesce behind him. And in an appeal to the centrist independents who will decide the general election, Romney pledged Tuesday to work with Democrats or "die trying."
"Tonight was a primary, but November is a general election. And we're going to face a defining decision as a people," Romney said during a victory speech to supporters. "We know what Barack Obama's vision is. We've been living it these last three years. My vision is very, very different."
Romney picked up at least 41 delegates in Illinois, according to initial results, adding to his delegate lead and making it that much harder for any of his rivals to deny him an opportunity to take on the president in November.
Obama, for his part, was headed West on Tuesday to Nevada, New Mexico and Oklahoma City on a trip aimed at answering critics of his energy policies, sure to be a key issue in the fall campaign. His first stop was a plant in Nevada that uses solar panels to power homes, part of an effort to highlight the president's programs to expand renewable energy sources.
Romney was moving on to Maryland, where polls show he has the advantage heading toward an April 3 primary.
With Louisiana voting on Saturday, the South could prove less hospitable to Romney.
Santorum, who hopes to rebound in Louisiana, sounded like anything but a defeated contender Tuesday night as he spoke to supporters in Gettysburg, Pa. He said he had outpolled Romney in downstate Illinois and the areas "that conservatives and Republicans populate."
"We're very happy about that and we're happy about the delegates we're going to get, too," he said before invoking Illinois-born Republican icon Ronald Reagan, the actor turned president. "Saddle up, like Reagan did in the cowboy movies."
Former House Speaker Newt Gingrich didn't speak to supporters Tuesday, instead putting out a written statement. Texas Rep. Ron Paul has yet to win a state.
Romney triumphed in Illinois after benefiting from a crushing 7-1 advantage in the television advertising wars, and as his chief rival struggled to overcome self-imposed political wounds in the marathon race to pick an opponent to Obama.
Most recently, Santorum backpedaled after saying Monday that the economy wasn't the main issue of the campaign. "Occasionally you say some things where you wish you had a do-over," he said later.
Romney has 563 delegates in the overall count maintained by The Associated Press, out of 1,144 needed to win the nomination. Santorum has 263 delegates, Gingrich 135 and Paul 50.
After the Louisiana primary, a 10-day break follows before Washington, D.C., Maryland and Wisconsin hold primaries on April 3.
Santorum is not on the ballot in the nation's capital. Private polling shows Romney with the edge in Maryland, and the pro-Romney super PAC Restore Our Future launched a television ad campaign in the state during the day at a cost of more than $450,000.
Wisconsin shapes up as the next big test between Romney and Santorum. Republican politics there have been roiled recently by a controversy involving a recall battle against the governor and some GOP state senators who supported legislation that was bitterly opposed by labor unions.
Already, Restore Our Future has put down more than $2 million in television advertising across Wisconsin. Santorum has spent about $50,000 to answer.
Neither Gingrich nor Paul campaigned extensively in Illinois. Gingrich has faded into near-irrelevance in the race, but he remains defiant.
"To defeat Barack Obama, Republicans can't nominate a candidate who relies on outspending his opponents 7-1," he said in a statement Tuesday night. "Instead, we need a nominee who offers powerful solutions that hold the president accountable for his failures."
Gingrich said his campaign will spend the time leading to the party convention "relentlessly taking the fight to President Obama."

Leading thinkers in agriculture, climate change and the environment will gather in Nairobi, Kenya this April to review innovative ways to tackle Africa's unending cycle of drought and food insecurity. The Beating Famine conference comes as yet another food crisis sweeps across the Sahel region of Africa, this time through Niger, Mali, Burkina Faso, Chad and Mauritania.
The conference will take place on April 10-13 at the World Agroforesty Centre in Nairobi. A joint initiative by World Vision and the World Agroforestry Centre, the forum will bring together policy-makers from across Africa, leading agriculture/food security/environment experts, international NGOs, donors, academia, practitioners and the media.
"The world watched as millions suffered from famine in the Horn of Africa last year and now that suffering is spreading to parts of West Africa," said conference organiser Rob Francis. "These crises are calling for a long-term sustainable approach to food insecurity and famine, and we believe the answer lies with a greater emphasis on the environment and better agricultural practices."
The conference will focus on practical, low cost and proven techniques to reverse land degradation and deforestation, lift incomes, adapt and mitigate against climate change, and ultimately prevent famine.
Dennis Garrity, UN Drylands Ambassador and senior fellow at the World Agroforestry Centre noted that "The national planning segment of the conference will be particularly significant. Governments and NGOs throughout the East African region are now partnering to implement national scaling-up programmes to create an evergreen agriculture based on smallholder adoption of trees for enhanced soil fertility, and fodder, fruit and fuelwood production. Accelerating the widespread use of these practices would have an enormous impact."
"In essence, we hope to spark a regreening movement that transforms thinking across the world," said World Vision East Africa climate change and environment specialist Assefa Tofu. "Governments, development organisations and also the community must learn to see the power of simple, effective environmental techniques as a new way of tackling hunger."
The conference will consist of presentations by high level delegates, as well field trips and demonstrations. Participants will also be encouraged to develop action plans for their country or region.
A demonstration of Farmer Managed Natural Regeneration (FMNR) will be conducted by FMNR pioneer Tony Rinaudo on Friday April 13 in Kijabe, Kenya. FMNR has helped make great advances for the food security and economic sustainability of farmers in eight countries across Africa and three in Asia.
